Secreted markers for osteoblast and osteoclast function in 2D and 3D cocultures of THP-1 cells and SaOS-2 cells. THP-1 cells and SaOS-2 cells (2:1 ratio) were cocultured over 14 days in both 2D and 3D on pHEMA-based cryogel scaffolds. On days 1, 4, 7, 10, and 14 of culture, (A) soluble receptor activator of nuclear factor kappa-Β ligand (sRANKL), (B) osteoprotegerin (OPG), and (C) the sRANKL:OPG ratio, (D) procollagen type I N-terminal propeptide (PINP), (E) collagen-type I N-telopeptide (NTX), and (F) osteocalcin levels were detected in the culture supernatant. Data are presented as curves (mean ± SEM).
