COX-2 expression induced by NO-donors. (A) HUVEC were incubated for 18 h with different concentrations of SNAP, GSNO, SNP alone or in combination with IL-1α. (B) The generation of peroxynitrite was monitored by the fluorescence measured at Ex/Em = 490/530 nm using the peroxynitrite assay kit (C) HUVEC were preincubated with 100 μM uric acid for 1 h. Incubation was continued for 18 h after the addition of 1 mM SIN-1. COX-2 expression was evaluated by Western blot analysis. β-actin was used as a control of protein loading.
