Granulosa cells from WT and Sod2+/− mice were isolated and cultured as described in Materials and Methods. Granulosa cells were plated at a density of 1 × 105 cells/well and incubated with medium alone, FSH (100 ng/ml), Bt2cAMP (2.5 mM), hHDL3 (500 μg protein/ml), FSH + hHDL3 or Bt2cAMP + hHDL3 for 48 h. Media were collected for analysis of progesterone and 20α-hydroxyprogesterone by specific RIAs. Results are Mean ± SE of four independent experiments.
