Granulosa cells from WT and Sod2+/− mice were isolated and cultured as described in Materials and Methods. Granulosa cells were plated at a density of 1 × 105 cells/well and incubated with hHDL3 (500 μg protein/ml), FSH (100 ng/ml) + hHDL3, or Bt2cAMP (2.5 mM) + hHDL3 for 48 h. Media were collected for analysis of estradiol by using an ELISA kit (Sigma-Aldrich). Results are Mean ± SE of four independent experiments. *p < 0.05; **p < 0.01.
