(A) BEND3-knockout OCI-AML2 cells (1 × 106) were injected subcutaneously into the flanks of SCID mice. When the tumors became palpable, mice were randomly divided into 5 groups (n = 10 per group) and treated with vehicle (10% HPBCD in water), TAK-243 (10 or 20 mg/kg), Ko143 10 mg/kg, or a combination of TAK-243 10 mg/kg + Ko143 10 mg/kg subcutaneously twice weekly for 3 weeks. Asterisks shown denote significantly different final tumor volumes in treated groups compared with vehicle, determined using repeated-measure 2-way ANOVA and Sidak’s multiple comparisons test. (B) After 3 weeks, mice were euthanized and tumors harvested and weighed. Significance of difference was determined using 1-way ANOVA and Tukey’s multiple comparisons test. (C) Images of tumors harvested from the 5 groups are shown. (D) Mice were weighed every 2–4 days. Data points (A, B, and D) represent means ± SEM. *P ≤ 0.05; ****P ≤ 0.0001.
