Correlation between bacterial burden and induction of various nutritional reporters. HeLa cells (A, B) or RAW264.7 macrophages (C, D) were infected with STM WT harboring p5008 [PEM7::DsRed PsitA::sfGFP], p5069 [PEM7::DsRed PsufA::sfGFP], p5067 [PEM7::DsRed PbioA::sfGFP], p5078 [PEM7::DsRed PcorA::sfGFP], p5003 [PEM7::DsRed PznuA::sfGFP], or p5080 [PEM7::DsRed PkdpA::sfGFP] as indicated at MOI 5. The cells were detached and fixed 16 h p.i. Subsequently, host cells were analyzed by FC to distinguish non-infected (gray frame), from infected cells by virtue of bacterial DsRed fluorescence. At least 10,000 infected cells per condition were analyzed and gated for host cells harboring sfGFP-negative (black frame) or sfGFP-positive STM (green frame). Data for STM WT in HeLa cells (A, B) or RAW264.7 macrophages (C, D) of a representative experiment from three biological experiments are shown.
