Changes in intracellular biotin availability for STM in HeLa cells or RAW264.7 macrophages. HeLa cells or RAW264.7 macrophages were infected at MOI 5 with STM WT, ΔssaV or ΔsifA as indicated, each containing the biotin reporter p5067. 1 mM D-biotin (orange) was added 1 h p.i. to STM-infected HeLa cells or RAW264.7 macrophages, and maintained for the rest of the infection period. Host cells were lysed 16 h p.i., released STM were fixed, and subjected to FC to quantify sfGFP intensities of PbioA-positive STM. Data for STM WT [p5067] in HeLa cells (A) or RAW264.7 macrophages (C) of a representative experiment are shown. Representative quantification of X-mean sfGFP intensities for STM WT, ΔssaV and ΔsifA in HeLa cells (B) and RAW264.7 macrophages (D) at 16 h p.i. are shown. Mean values and standard deviations of PbioA-positive bacterial populations from triplicates of a representative experiment from three biological replicates are shown.
