CcaF1 affects stability of some coding and non-coding RNA transcripts. Determination of RNA half-life of R. sphaeroides wild type and wild type with pRKCcaF1 for selected coding and non-coding RNA transcripts. Cells were harvested in exponential growth phase under microaerobic conditions. Samples were taken after adding rifampicin at different time points. Total RNA was isolated and either run on a 10% denaturing polyacryamide gel (upper six panels) or on a 1% formaldehyde agarose gel (lower four panels) and blotted. 5S or 14S rRNA serve as loading controls. For quantification RNA signal intensities were normalized to 5S or 14S rRNA signals. The average half-life was calculated from three independent experiments and the standard deviation is indicated.
