Reads were generated in an E1 in vitro selection experiment conducted using an E1-randomized library with a ‘GG’ IG background. Results are presented in both tabular and graphical forms. (B) List of the 10 most frequently represented E1 variants among qualifying sequence. (C) Sequence composition and results obtained from synthetic PIE constructs containing native E1 and IG sequences (WT,WT), native E1 and ‘GG’ IG (WT, GG), the two best-represented variants isolated in the in vitro selection experiment (s1a, GG; s1b, GG), or these variants plus compensatory mutations in IG (s1a, GGc; s1b, GGc). Constructs were processed and analyzed as described for Figures 3 and 6.
