HEK293T cells expressing the components of the BRET biosensor for Gαi-GTP, the membrane-anchored FRB construct, M4R, and FKBP-fused GIV GBA or R12 GL were sequentially stimulated with carbachol (100 μM) and rapamycin (0.5 μM) at the indicated times during kinetic BRET measurements.
