PRSS1 is a direct target gene of miR-146a-5p. (A) The TargetScan database was used to predict the binding site of miR-146a-5p to PRSS1. (B) Luciferase assay of HEK 293T cells cotransfected with miR-146a-5p mimic, miR-146a-5p mimic NC and a luciferase reporter containing the wild-type 3′UTR of PRSS1 (PRSS1-3′UTR-wt) or mutated miR-146a-5p binding sites on the 3′UTR (PRSS1-3′UTR-mut). (C-D) qRT-PCR was performed to detect the expression changes of miR-146a-5p and PRSS1 mRNA in MGC803 cells transfected with miR-146a-5p mimic and miR-146a-5p inhibitor, respectively. (E) The effect of miR-146a-5p mimic or miR-146a-5p inhibitor on PRSS1 protein expression in MGC803 cells was determined by Western blot.
