Effects of antibiotic treatment on bacterial load in organs 24 h upon injection of BMDM infected with S. aureus. BMDM of (A) wildtype or (B) nox2−/− mice were infected with S. aureus MW2 GFP (MOI30). 2.5 × 106 infected BMDMs were injected in PBS i.v. into naive C57Bl6J mice (n=6 per genotype and treatment). (C) Mice injected with 4 × 106 plain S. aureus MW2 GFP, corresponding to the CFU recovered from infected BMDM (n=6 per treatment). 200 mg/kg vancomycin was injected s.c. either alone or in combination with 10 mg/kg Rifampicin at 1 h, 6 h, 24 h, and 30 h after i.v. infection. At 48 h p.i. livers of infected mice were homogenized and plated on Mueller-Hinton plates. Shown are individual values for each liver in CFU/g. Red line indicates median, dashed line represents limit of detection. Student's t-test was performed on log-transformed values.
