Characterization of the thermal stability and tolerance to organic solvents for RsPtxD variants. (A) Thermal inactivation of RsPtxD mutants at 45°C. Prior to the assay reaction, RsPtxD mutants were incubated in the 50 mM MOPS (pH7.3) containing 0.5 mM NAD (filled circles), 0.5 mM NADP (open circles), or without cofactor (squares). The reactions were performed by using 0.5 mM NADP as a nicotinamide substrate. The data are representative of two independent experiments.
