Developmental sensitivity to TSA depends on a functional TTD of Sgf29. (A) Schematic illustration of wildtype and F359A/Y366A Sgf29 expression constructs. The expression of C-terminally 3xFLAG-tagged protein is driven by the endogenous promoter of sgf29 in an integrating vector. (B) Expression of Sgf29–3xFLAG in sgf29 cells re-sensitizes cells to TSA during development. Upper panel, immunoblots showing expression of 3xFLAG-tagged wildtype and F359A/Y366A Sgf29 in sgf29 cells. Whole cell lysates control sgf29 cells and sgf29 cells expressing wildtype or F359A/Y366A Sgf29–3xFLAG was resolved by 12% SDS-PAGE and blotted using anti- FLAG antisera. Actin is used as a loading control. Lower panel, cells were washed with KK2 and allowed to develop on buffered agar with increasing concentration of TSA as described in Figure 1A. Images were taken at 24 h and representative images of three repeats shown. The percentage of late structures is shown as average ± SD (n = 3).
