In vitro effect of combined treatment with hTERT-ADSC.sTRAIL and cpt-11. (A) The suicide effect. At greater than 1 μM CPT-11, the viability of ADSC.sTRAIL cells was lower than at more than 0 μM CPT-11 (P < 0.05). (B) The cytotoxicity of ADSC.sTRAIL and 5 μM CPT-11 toward PC3 at cocultured ratios of 0.05. The cell viability of PC3 decreased significantly by treatment with both CPT-11 and ADSC.sTRAIL compared with CPT-11 treatment alone. (D) Flow cytometry for propidium iodide/annexin V staining showed that the percentage of annexin V–positive apoptotic PC3 cells increased in the presence of ADSC.sTRAIL and CPT-11 compared to CPT-11 treatment alone. (E) The percentage of apoptotic cells significantly increased in the ADSC.sTRAIL-treated group compared with those treated with cpt-11 alone (P < 0.05).
