Protein kinase A (PKA)-dependent phosphorylation of proteasomal subunits at the onset of human sperm capacitation. (B) Spermatozoa were incubated under capacitating conditions for 0 or 15 min with 0.1% v/v DMSO or for 15 min with 50 μM H89. Then, the proteasome was immunoprecipitated with an anti-PSMA7 proteasome subunit antibody. The phosphorylation of proteasomal subunits by PKA was evaluated by western blots using an antibody against phosphorylated PKA substrates. (B) For the densitometric analysis, the PSMA7 subunit was used as a control. The bars represent the mean ± SEM of total signal from phosphorylated proteosome components. N = 4.
