IL-35 expression differed in lungs and spleens of different ARDS models. C57BL/5 mice (5/group) were subjected to LPS or CLP. (A, B) IL-35 mRNA levels in the lungs were measured with qRT-PCR. Relative expression levels of the genes were expressed with the GAPDH housekeeping gene as an internal reference. (C–J) Organs were removed at the indicated time points, Blood specimens were collected from mice under anesthesia via the ophthalmic vein. Bronchoalveolar lavage fluid was obtained by washing the bronchus three times with 0.2 mL of sterile PBS each time, and the homogenate was obtained by mixing tissue and PBS in a ratio of 0.5 g:1 mL. Samples were assayed for IL-35 content by enzyme-linked immunosorbent assays.*p < 0.05, **p < 0.01, ***p < 0.001, and ****p < 0.0001, by the one-way ANOVA followed by LSD multiple comparisons test, compared with normal mice.
