Tubule formation and duration in Ap3d1−/−/Pldn−/− melanocytes stably expressing AP-3δmut1 and Pldnmut1 (related to Fig. 7). Ap3d1−/−/Pldn−/− melanocytes stably expressing AP-3δmut1/Pldnmut1 were transiently transfected with GFP-RAB11 and imaged 48 h later by live-cell spinning disk confocal fluorescence microscopy in a humidified chamber at 10% CO2 and 37°C. Images were acquired for 2 min at 1 frame/s. The yellow arrow indicates a GFP-RAB11–positive tubule that persists for 5 s.
