Different metal selectivity of SARS-CoV-2 nsp14-nsp10 exonuclease and RTC. (A) Metal selectivity of SARS-CoV-2 nsp14-nsp10. The nuclease activity of SARS-CoV-2 nsp14-nsp10 was assayed in the presence of EDTA or divalent metals. (B) Metal selectivity of SARS-CoV-2 RTC. SARS-CoV-2 RTC extends 20-mer primer to the full-length 40-mer product. In both nsp14-nsp10 and RTC, to make a “metal-free” enzyme state, EDTA (1 mM) was preincubated with nsp14-nsp10 and then excess divalent metals (2 mM) were added. The substrate in Fig. 1D was used for these experiments. Experiments were repeated twice and the representative assay is shown here.
