NF-κB inhibitor inhibited the hyperinduction of cytokines by SFTSV infection. (A) HEK293T cells were mock treated or infected with SFTSV at a MOI of 1 × 103 or 1 × 104 TCID50 in the absence or presence of TPCA-1 (20 μM). The expression of the IL-8, TNFA, CCL2, and viral nucleoprotein NP genes was quantified by qRT-PCR at 24 and 48 h of infection. (B) Fourteen Ifnar−/− mice were intraperitoneally inoculated with 2 × 106 TCID50 of SFTSV on day 0. On day 2, half of the mice were given 1 mg of TPCA-1 intraperitoneally, and the rest were given PBS as a control. On day 3, the spleen was harvested from each mouse and RNA was extracted, purified, and reverse transcribed. Then, expression of Cxcl1, Il-6, Ccl2, Tnfa, and viral nucleoprotein NP genes was quantified by qRT-PCR. Data are presented as means ± SEM. The Student t test was used for statistical analysis.
