FIG 3 — Adsorption and replication of phage T4 in JM109 cells. (A) Adsorption assay measured the percentage of extracellular free phage T4 remaining over a 25-min period. PT-based SspFGH defense in JM109(pWHU4321, pWHU4322) does not interfere with phage adsorption. (B) Southern blot analysis of the phage T4 genome in infected JM109 cells containing PT-based SspFGH or empty vectors over a 60-min period. The probe was designed to match region 148,913 to 149,433 in the T4 genome. Each lane contained 2 μg of total intracellular DNA digested with PacI or 1.5 μg of undigested total DNA. U, uninfected.
