LNPs with T7 pass through the transwell cell barrier and are taken up by target cells. HeLa (CCR5-negative control cell) or TZM-bl (CCR5-positive cell type) cells (A and B) as well as HEK293T or gp160 positive HEK293T cells (C and D) were seeded at a density of 50,000 cells/well. The next day, transwell inserts containing confluent hMEC/D3 cells at trans-endothelial electrical resistance (TEER) above 30 Ω⋅cm2 were placed into experimental wells, LNPs (1000:1) were added to the apical surface, and 24 h later, the target cells were processed for Cy5 detection using fluorescence-activated single-cell sorting (FACS).
