RNA from HepG2 transfection studies with wt and mutant HBV plasmids (G1738A, G1748A, and G1738A/1748A), harvested at days 1, 3, 5, and 7 posttransfection. RNA was extracted using Trizol extraction followed by DNase digestion. qPCR was performed using primer sets to pick up the core transcripts and total RNA (see Table S3) as noted in the schematic (A). The information is represented as a ratio of core transcripts to total RNA for both the supernatant (B) and cells (C) to account for equivalent detection of residual forms of DNA as detected by qPCR.
