TABLE 1.

Transformation and differentiation rates of pBDL23- and pBDL22-transformed rice calli.

Vector Initial calli Transformed calli * Transformation rates (%) Differentiated calli ** Calli differentiated into plants Differentiation rates (%) Plants transplanted to soil ***
pBDL23 562 378 67.26 232 128 55.17 73
pBDL22 557 333 59.78 212 103 48.58 79

*Nipponbare rice calli were co-cultured with Agrobacterium for 3 days and subjected to 19 days of selection culture on hygromycin medium. The pBDL23-transformed calli expressing GFP and mCherry fluorescence and the pBDL22-transformed calli expressing GFP fluorescence were counted separately. For each vector, the transformation rate is the percentage of the transformed fluorescent calli among the calli initially cultured on hygromycin selection medium. **Transformed fluorescent calli described as above were subjected to a second selection culture for 20 days, and then cultured on differentiation medium for 48 days to be regenerated into plants. For each vector, the differentiation rate is the percentage of the fluorescent calli that were regenerated into plants among the fluorescent calli initially cultured on differentiation medium. ***Healthy pBDL23-transformed plants expressing GFP and mCherry and pBDL22-transformed plants expressing GFP were transplanted into soil, and T1 seeds were harvested at maturity.
