IL-4 treatment increases viral replication in macrophages but does not reactivate virus from a latently infected B cell line. (A) Bone marrow-derived macrophages (BMDMs) were treated with 100 ng/ml IL-4 and 20 ng/ml PMA for 16 h in culture medium and then infected with MHV68 at an MOI of 5. Twenty-four hours after infection, cells were fixed, and cells expressing lytic viral proteins were determined by flow cytometry. (B) HE2.1 B cells were treated with 100 ng/ml IL-4 and 20 ng/ml PMA for 38 h, cells were then fixed, and cells expressing lytic viral proteins were determined by flow cytometry. SSC, side scatter.
