KD of Dsc2 in human model IECs resulted in reduced intercellular adhesion. (A) SKCO-15 cell monolayers with KD for Dsc2 vs. control NS were subjected to a dispase II-based mechanical cell dissociation assay to induce monolayer fragmentation as a readout of cell–cell adhesion. (B) Depletion of Dsc2 resulted in significantly increased epithelial monolayer fragmentation suggesting impaired cell–cell adhesion. Results show the mean  ±  SEM of data from three individual experiments, each one assayed in two technical replicates. Statistical analysis was done with two-tailed Student’s t test. **p < 0.01.
