AP collagen peptides prevent cortisol-dependent inhibition of collagen synthesis in a reconstituted human skin model. The model was treated with cortisol (100 nM) in the presence of APCP (1 μg/mL), GR inhibitor (1 μM) or TGF-β (10 ng/mL) and cultured for 48 h. (A) Reconstituted human skin models were fixed and stained with H&E, Masson’s trichrome (MT) or immunohistochemical staining for collagen type 1 antibody. Scale bar: 100 μm. (B) Total RNA was extracted from a reconstituted human skin model, and relative col1a1 mRNA was measured via qRT-PCR. Data represent the mean ± SD of two independent experiments. (C) Secreted procollagen type 1 was detected in culture supernatants using a specific ELISA kit. Data represent the mean ± SD of three independent experiments.
