NPG treatment increases ROS production in PDAC cells. PDAC cells were treated with NPG (GEM 0.5 M, nab-paclitaxel 0.1 M) for 24 h. Subsequently, cells were preloaded with 2′,7′-dichlorodihydrofluorescein diacetate (DCF) or dihydroethidium (DHE) and studied under a fluorescent microscope. (A,C) show Capan-1 controls, and (E) shows CD18/HPAF cells control cells treated with vehicle only. (B) shows NPG-treated cells loaded with DCF, whereas (D,F) show NPG-treated cells loaded with DHE. The panel on the right side shows fluorescence intensity of the control and NPG-treated cells. (p < 0.05, n = 3).
