Immunoproteasome activity recovers after pulse treatment with sub-toxic doses of ONX-0914. The combined activity of LMP7 and ß5c sites was measured with Suc-LLVY-amc in extracts of ONX-0914-treated cells harvested immediately after 1 h treatment or after subsequent culturing in drug-free media (n = 3–7). On the right panel, extracts were treated with the highly specific immunoproteasome inhibitor LU-015i immediately before activity measurements (n = 2–3).
