Expression of β-Klotho mRNA, Fgf21 mRNA and FGF21 protein in skeletal muscle from WT and Fgf2KO mice. Young (5 m) and old (20 m) WT and Fgf2KO mouse hindlimb gastrocnemius muscles were isolated and total RNA extracted for qPCR (a) β-Klotho mRNA; (b) Fgf21 mRNA were significantly increased in old muscle of both genotypes n = 4–6 mice/group. ^#Compared to corresponding 5 m p < 0.05 (c) Young (5 m) and old (20 m) WT and Fgf2KO mouse tibialis anterior muscles were isolated, sectioned and immunohistochemistry for FGF21 revealed increased FGF21 in muscle cells of old WT and Fgf2KO compared with young of both genotypes.
