Validation of the compartmental ODE model outcomes against published experiments. (A) The ratio of pFAK to total FAK as a function of stiffness was validated against data from Beamish et al. (51) for both normal human lung fibroblasts (NHLF) and renal proximal tubular epithelial cells (RPTECs) seeded on 2D PEG hydrogels of various stiffnesses and glass. (B) RhoA-GTP activation over stiffness model fit and comparison to experimental data from endothelial cells seeded on 2D polyacrylamide gels of various stiffnesses and glass (32). (C) Ratio of myosin activation over total myosin predicted by our model and a comparison with experimental data from endothelial cells seeded on 2D polyacrylamide gels of various stiffnesses and glass (32). (D) Validation of Lamin A activation as a function of tissue stiffness across various tissues including brain, kidney, and bone among others (43). (E) The ratio of nuclear YAP/TAZ to cytosolic YAP/TAZ as predicted by our model and a comparison with experimental data from multiple sources (17, 46–48) (six utilized 2D PA gels and eight utilized 2D hyaluronic acid gels) (6, 8, 17, 46–48). (F) Sensitivity analysis with respect to YAP/TAZ Nuc/Cyto ratio for 3.5 kPa. Additional sensitivity analyses for all of the variables to all of the parameters and all initial conditions can be found in SI Appendix, Supplemental Methods (SI Appendix, Figs. S4 and S5).
