CHX triggers the degradation of PM, but not VM proteins, related to Fig. 3. Mid-log cells expressing either a PM-GFP protein (Mup1-GFP or Can1-GFP) or a VM-GFP protein (Fet5-GFP, Fth1-GFP, or Vph1-GFP) were mixed at a 1:1 ratio before being loaded into a microfluidic imaging chamber. After a brief recovery (30 min), the CHX-containing medium was perfused into the imaging chamber (time 0). Cells were incubated in the CHX-containing medium for 3–4 h, and images were taken every 30 min. The video plays at 2 frames per second.
