Heat triggers the degradation of PM, but not VM proteins, related to Fig. 3. Mid-log cells expressing either a PM-GFP protein (Mup1-GFP or Can1-GFP) or a VM-GFP protein (Fet5-GFP, Fth1-GFP, or Vph1-GFP) were mixed at a 1:1 ratio in a shaking flask and diluted to OD600 = 0.10. The flask was then placed at 37°C for another 3–4 h. Samples were collected every hour and analyzed by snapshot imaging. PM-GFP strain was colabeled with Vph1-mCherry. Z stacks (0.4 µm per step) of the uncropped original images were shown at each time point. The video plays at 2 frames per second.
