NLRC5 binds to and activates NLRP3/NLRC4 inflammasomes during RIR or OGDR. (A–F) Western blot and RT-qPCR analysis of NLRP3 and NLRC4 expression in BV2 microglial cultures and mouse retina following OGDR and RIR, respectively. Expression levels of both proteins and mRNAs were elevated in vivo and in vitro by RIR or OGDR. Elevated expression levels following OGDR were markedly suppressed by NLRC5 knockdown. (G,H) Co-immunoprecipitation assay showing that NLRC5 interacts with NLRP3/NLRC4 in BV2 cells subjected to OGDR.
