Generation of NK cell-specific IL-18 receptor-deficient mice. Excision of Il18r1 allele occurs predominantly in NK cells of NKp46-Cre-Il18r1fl / fl mice. (A) Protein expression of the IL-18 receptor alpha (IL-18Rα) on different subsets of splenic leukocytes from indicated mice. (B) NK cells from spleens of naïve Il18r1fl / fl (Cre-) and NKp46-Cre-Il18r1fl / fl (Cre+) littermate mice were enriched and stimulated with IL-18 ex vivo for 24 hours. 100 U/ml of rhIL-2 was added to maintain NK cell survival. Representative plots depict the mean fluorescence intensity (MFI) of CD71 and CD98 expression on cytokine-stimulated NK cells. The MFI expression is presented in percentage relative to the MFI of unstimulated cells as 100. Data are from one experiment representative of 3 independent experiments, with at least two replicates per group. Data represent mean + SD. NS, non-significant; ***p < 0.001.
