Fig 6. Mutation of ObDMR6 in T2 complete knockout mutants enhances resistance to Peronospora belbahrii.

Fig 6

(a) Pathogen biomass determined by qPCR. Infected leaf samples were collected from the wild-type (WT) and T2 plants derived from indicated T1 lines at 4 days post inoculation (dpi). The pathogen biomass was quantified as the ratio of amplification of P. belbahrii ITS2 relative to O. basilicum β-tubulin. Values represent mean ± standard error of three biological replicates with each consisting of two technical replicates. (b) Production of sporangia on inoculated leaves at 9 dpi, calculated as the number of sporangia per gram of fresh weight. Values represent mean ± standard error of three biological replicates. Statistically significant differences between the mutants and WT were determined by one-tailed t test, with one asterisk (*) indicating P < 0.05 and two asterisks (**) indicating P < 0.01. (c) Differential basil downy mildew disease levels of WT and T2 plants derived from indicated T1 lines shown with sporulation on the abaxial side of the leaves at 9 dpi. The experiments were repeated three times with similar results.
