Construction of BC3242 IgA1 HC. The sequence of the ZAC3 Vibrio cholerae antibody heavy chain was obtained from published sequences. The sequence was reverse translated, the DNA optimized, and synthesized by GeneArt, (Thermo Fisher). The HC sequence was obtained as a SalI 1.5kb fragment in the mammalian expression vector pcDNA3—3238 HC. This SalI fragment was isolated and ligated into the XhoI site of the beta casein expression vector BC451 to yield BC3242 IgA1 HC.
