Figure 2

Target-based whole-cell screening of compounds 1i and 1f against a coaBC Tet-OFF hypomorph. The impact of ATc-mediated transcriptional silencing of coaBC on the relative growth of a coaBC hypomorph (a,b; d,e) and wild-type Mtb H37RvMA (c) in the presence of increasing concentrations of 1i (a), 1f (b, c), rifampicin (d), and isoniazid (e) was determined. Data are representative of the mean and SD of independent triplicates. ATc, anhydrotetracycline; Tet-OFF, mutant whose growth is inhibited in the presence of ATc; PantS2.5, pantethine (2.5 mg/mL); RIF, rifampicin; INH, isoniazid.
