Figure 3

Exosomes inhibited LPS-induced alveolar macrophage viability loss and apoptosis. (a) The optimal action concentrations of LPS on alveolar macrophages were tested by Cell Counting Kit-8 assay. (b) The protective effects of BMSC-derived exosomes on alveolar macrophage viability were estimated by Cell Counting Kit-8 assay. (c) The apoptosis observed by the TUNEL assay in alveolar macrophages. The fluorescent signals from fragmented DNA (green) and nuclei (blue) were visualized and photographed under an inverted fluorescence microscope (×400 magnification). Scale bar represents 50 μm. (d) The apoptosis rate of alveolar macrophages was performed by flow cytometry with Annexin V-FITC/PI double staining. (e) The percentages of apoptotic cells are shown in the bar chart. Values are mean ± SD of three experiments each carried out in triplicate. P < 0.05 vs. the control group; ∗∗P < 0.01 vs. the control group; #P < 0.05 vs. the LPS group; ##P < 0.01 vs. the LPS group.
