The retromer and retriever complexes. The retromer recognition core Vps35–Vps29–Vps26 trimer (1) recruits subcomplexes, such as the SNXs‐BAR (a) and WASH (b), and Rab7a (c) and SNARE (d) proteins to promote cargo (C) sorting. Targeted cargoes, including cation independent mannose 6‐phosphate receptor (CI‐MPR; e), sortilin‐related receptor (SorLa; f), and amyloid‐β (Aβ) precursor protein (APP; g), are transported from the endosome to the trans‐Golgi network (TGN) by the retromer complex. The retriever complex (DSCR3–C16orf62–VPS29) (2), associated with sorting nexin 17 (SNX17) protein, WASH, and CCC complexes, cooperates with the retromer complex to mediate the endosomal protein sorting process.
