Preparation of Hungate tube

Seal Hungate tube using a butyl rubber septum, followed by a screw cap in an anaerobic chamber. Sterilize the sealed Hungate tube by autoclaving. Following transport of the Hungate tube into an anaerobic chamber, transfer the appropriate solution into the Hungate tube using a sterile syringe and needle.
