Alkaline pH sensitivity of B. thailandensisdbcA. (a) Growth rate of B. thailandensis strains (E264; vec, E264; DbcA, ∆dbcA; vec, and ∆dbcA; DbcA) at different times in Mueller Hinton broth (MH1) with 100 µg/ml Tmp. The error bars indicate standard deviations of three biological replicates. (b) Spot assay of E264; vec, E264; DbcA, ∆dbcA; vec, and ∆dbcA; DbcA on MH1 agar media. 1:10 dilutions of indicated strains were spotted on NaOH adjusted MH1 media with 100 µg/ml Tmp and 0.001% rhamnose. Plates were analyzed after 24 or 48 h at 37 °C. (c) Comparison of alkaline pH sensitivity of B. thailandensisdbcA in MH2 with MH1 agar media. MH2 contains ~ 20 to 25 mg/l of calcium and ~ 10 to 12.5 mg/l of magnesium, whereas MH1 contains no added calcium and magnesium salts. The pH was adjusted by either HCl or NaOH. Plates were analyzed after 48 h of growth at 37 °C.
