Schematic representation of the protocol to process and infect fresh human skin explants. After removal of the adipose tissue, the skin explants were pinned in a dissection board and the surface of epidermis was tape-stripped with an hypoallergenic tape. Skin punches of 8 mm were prepared, washed in PBS and placed in a transwell, previously filled with culture medium. The skin was infected with 5 × 10⁶ CFU of USA300 LAC strain and cultured at air-liquid interface for 2, 3, 24, or 72 h.
