Inhibitory pathway of colchicine. Colchicine inhibits activation of purinergic P2X2/P2X7 receptors and blocks cation uptake and subsequent pro-inflammatory signaling cascades without affecting cell death. Colchicine also inhibits the NALP3 inflammasome pathway, the Rho/ROCK pathway via cytoskeleton rearrangement, and inhibits release of ROS, NO and TNFα.
