SARS-CoV-2 protein expression in HeLa cells. (A) Genome structure of SARS-CoV-2. The ORF1a/1ab encodes non-structure proteins, four genes encode four structural proteins (S, E, M, N), and six accessory genes encoding six accessory proteins (ORF3a, ORF6, ORF7a, ORF7b, ORF8, and ORF10) are indicated. (B) SARS-CoV-2 ORF3a, M, 7a, N genes were fused with a FLAG-tag (F) at the N-terminus and cloned in the pXJ41 expression vector. pF-ORF3a, pF-M, pF-ORF7a, and pF-N were transfected in HeLa cells for 24 h, and cell lysates were subjected to immunoblot using α-FLAG PAb. Beta-actin served as a loading control. The full-length blot is presented in Supplementary Fig. S1. (C) Distribution of SARS-CoV-2 ORF3a, M, ORF7a, and N protein in HeLa cells. Cellular localization of the individual protein (red) and cell nuclei (blue) were examined by confocal microscopy.
