Reaction scheme of split‐intein‐mediated trans‐splicing (PTS) of protein N termini. For split‐intein‐based techniques applied to the protein N terminus, the C‐terminal fragment (C‐Int, purple) is fused to the target protein (POI, gray), while the N‐terminal fragment (N‐Int, yellow) is artificially generated to carry the required reporter unit (green with red star). Split‐intein‐mediated PTS is a traceless labeling approach as the intein fragments are removed from the POI during the ligation of the exteins.
