Time-kill kinetics assay. (A) Time-kill curves of A. baumannii 17978 in blood after treatment with pepdD2 and pepI2. This experiment was performed in ex vivo whole rat blood at room temperature without shaking. A. baumannii (final concentration of 10⁷ CFU/mL) was incubated with or without peptide (4 μg/mL) for 60 min. At the indicated time points, an aliquot of the sample was removed and diluted, and the remaining cell number was counted by plating on MHA plates. The data are expressed as the mean ± SD. (B) The time-kill curves of A. baumannii 17978 after treatment with pepI2 and colistin sulfate in MHB. A. baumannii (final concentration of 10⁷ CFU/mL) was incubated with or without peptide at 37°C with shaking. The peptide concentration was 4 μg/mL for both peptides.
