Inhibiting mTOR-promoting autophagy and apoptosis in RMS cells of overexpressing GEFT. (A) Western blotting was used to detect the expression of Beclin1 and LC3 protein in RH30 cells transfected with GEFT, GEFT+Rapamycin, GEFT+CQ, and GEFT+CQ+ Rapamycin. (B) Western blotting was used to detect the expression of Beclin1 and LC3 protein in RD cells transfected with GEFT, GEFT+Rapamycin, GEFT+BafA-1, and GEFT+BafA-1+Rapamycin. (C) The representative images of RH30 and RD cells transfected with GEFT and GEFT+Rapamycin were detected by immunofluorescence, MDC and AO staining. (D) The apoptosis of RH30 and RD cells transfected with GEFT and GEFT+Rapamycin was detected by TUNEL. (E) Western blotting was used to detect the expression of mTOR, Bax, Bcl-2, Caspase-3, and Cleaved-PARP protein in RH30 and RD cells transfected with GEFT and GEFT+ Rapamycin. (F) Flow cytometry was used to detect the apoptosis of RH30 cells and GEFT-transfected RH30 cells in GEFT, GEFT+CQ, GEFT+ Rapamycin and GEFT+ Rapamycin +CQ groups.
