Tag-seq profiling the off-target sites induced by SpCas9 with 31 sgRNA in a single transfection. HEK293T cells were transfected with the donor Tag-oligo, SpCas9 and a pool mix of 31 sgRNA. Cells were harvested three days after transfection, and genomic DNA was extracted for library construction and then sequenced. All the off-target cleavages of the 31 sites can be parallelly profiled by Tag-seq.
