C2C12 cells were infected with lentiviruses expressing shRNA for scrambled (SC) or LPCAT3 (KD) sequences and differentiated into myotubes. (A) Caveolin-3 (cav3) protein content. (B and C) Detergent-resistant membranes (DRMs; fractions 4 and 5) and detergent-soluble membranes (DSMs; fractions 8–12) were isolated, and lipids were extracted for quantification of sphingomyelin (B) and cholesterol (C) (n = 4). (D) Cav3, Na/K-ATPase, actin, and total protein content were assessed via Western blot in all fractions from the sucrose gradient (image is representative of 4 experiments).
